5-benzoil-4-fenil-1H-primidin-2-on ve 5-benzoil-4-fenil-1H-primidin-2-tion bileşiklerinin UV-spektrofotometrik yöntemle kimyasal davranışlarının incelenmesi

Özet:

Bu çalışmada yeni sentezlenmiş iki primidin bileşiği; 5-benzoil-4-fenil-1H-primidin-2-on, (I), ve 5-benzoil-4-fenil-1H-primidin-2-tion, (II), nin, pH 1 -13 aralığındaki kimyasal davranışları incelenerek asitlik sabitleri tayin edilmiştir. Tayin işlemi Ultraviyole-görünür bölge spektrofotometri yöntemi kullanılarak gerçekleştirilmiştir. Çalışılan bileşiklerin proton bağlama ve proton serbest bırakmalarını gösterir bir mekanizma önerilmiştir. Her iki bileşiğin 1 pozisyonundaki azot atomuna bağlı hidrojen atomlarının çok asidik pH de dissosie oldukları gözlenmiştir. Diğer taraftan yine her iki bileşiğin 3 pozisyonundaki azot atomlarının da nisbeten daha yüksek bir pH de proton bağlama - proton salma reaksiyonu verdikleri bulunmuştur. I bileşiği ile ilgili olarak pH = 1.22 ve 7.49 da olmak üzere iki pKa değeri bulunurken, II bileşiği ile ilgili olarak sadece pH = 4.94 de bir pKa değeri bulunmuştur. I bileşiği ile ilgili olarak elde edilen ilk pKa değerinin 1 pozisyonundaki azot atomuna bağlı hidrojen atomunun dissosiasyonuna ait olduğu düşünülmüştür. Söz konusu hidrojen atomu asidik olduğundan birinci pKa değerinin üstündeki pH ler de dissosie olmaktadır. Bu dissosiasyon sonunda geride kalan azot atomu negatif yüklenmektedir. Yük dengesini korumak için çözelti ortamındaki bir katyonunun, muhtemelen lityum iyonunun buraya bağlandığı düşünülmüştür. II numaralı bileşiğin aynı pozisyondaki hidrojen atomu için maalesef bir pKa değeri bulunamamıştır. Bu farklılık bizim çalışma bileşiklerimize pilot model oluşturması nedeniyle üre ve tioürenin pKa değerleri göz önüne alınarak aşağıdaki şekilde açıklanmıştır: Üre ve tioürenin pKa değerleri sırası ile 0.1 ve -0.1 dir. Bu durumda, bizim II numaralı bileşikle ilgili olarak bulmamız gereken pKa değerinin de negatif bir değer olması beklenir. Bizim pH çalışma aralığımız 1-13 olduğundan, bu pKa değerinin tayin edilemediği düşünülmüştür. I bileşiği ile ilgili olarak elde edilen ikinci pKa değeri ile II bileşiği ile ilgili olarak elde edilen pKa değerinin 3 pozisyonundaki azot atomunun proton bağlama ve proton serbest bırakmasına ait olduğu düşünülmüştür. Yani bu azot atomları söz konusu pKa değerinin altındaki bölgede ortamda protonlanmış bir şekilde bulunurken, bu pKa değerinin üstündeki bölgede ise ortamda nötral bir halde bulunmaktadırlar. Deneysel çalışmalar hacimce % 5 metanol içeren bir metanol-su ortamında 25 ± 0.1 °C de yapılmıştır. Ortamın iyonik şiddeti LiCl ile 0.1 M olacak şekilde sabit tutulmuştur. SUMMARY In this study, chemical behaviors of recently synthesized 5-benzoyl-4-phenyl-1H-pyrimidine-2-one, (I), and 5-benzoyl-4-phenyl-1H-pyrimidine-2-thione, (II), have been investigated to determine their acidity constants, in the pH range 1 - 13. UV/Vis spectrophotometric technique was used in this study. A mechanism for the protonation and deprotonation pathway was proposed. The hydrogen atoms on the nitrogen atom at position 1 of both compounds were found to undergo deprotonation at lower pH values. However, the nitrogen atoms of both compounds at position 3 were found undergoing protonation and deprotonation reactions at relatively, higher pH values. Two pKa values were obtained for the compound I, at pH values 1.22 and 7.49. However, only one pKa value at pH = 4.94 was obtained for compound II. The first pKa value obtained for compound I was attributed to the deprotonation of the hydrogen atom which was bound to the nitrogen atom at position 1, in the pyrimidine ring. Since this hydrogen atom is acidic, it dissociated at pH values below the first pKa value. This dissociation caused the nitrogen atom to be charged negatively. Therefore, a positively charged ion,i.e. a cation probably lithium cation may balance the charge. For the compound II, however, no pKa value was obtained for the hydrogen atom which was bound to the nitrogen atom at position 1. This differency was explained as below; by taking into account the pKa values of urea and thiourea, which are a pilot model to our working compounds, I and II respectively. The pKa values for urea and thiourea are 1.0 and - 0.1 respectively. This knowledge indicated that the first pKa value for the compound II should be at minus pH values. Therefore, we could not determine it in the case of that we have studied the pH range from 1 to 13. The second pKa value (7,49) obtained for the compound I and the pKa value obtained at pH 4.94 for the compound II were attributed to the protonation or deprotanation reactions of the nitrogen atom at position 3, in the pyrimidine ring of both compounds. Namely, the nitrogen atom at position 3 of both compounds was existed in a protonized form at pH values below the second pKa values. However, they existed in a neutral form at pH values above the second pKa values. The experiments were performed in solutions prepared at a ratio of 5 % (by volume) methanol-water mixtures, at 25 ± 0.1 °C. The ionic strength of the medium was kept constant at 0.1 M with LiCl.
