Özet:

GENETİK ABSANS EPİLEPSİ (GAERS) DENEYSEL MODELİNDE TALAMUS VE KORTEKS PROTEOMLARININ PROFİLLENDİRİLMESİ Epilepsi beyindeki nöronların anormal aktivasyonları sonucu oluşan tekrarlayan nöbetlerle karakterize edilen kronik bir hastalıktır. Yaygın ve ciddi bir nörolojik hastalıktır. Prevalansı multipl sklerozdan 10 kat yüksektir ve dünya nüfusunun %0.5’inden fazlasını etkiler. Epilepsi üzerinde uzun süredir gerçekleştirilen çalışmalara rağmen, epilepsiye yol açan moleküler mekanizmalar tam olarak anlaşılamamıştır. Her ne kadar Na+, K+, Ca2+ kanal proteinleri ile ilgili genlerde mutasyonlar bildirilmişse de, hücre içi protein değişikliklerini gösteren çok az sayıda çalışma vardır. İki boyutlu elektroforez ve kütle spektrometresinin birleşimine dayalı proteomik tekniği evrensel olarak kullanılan bir tekniktir. Çözünebilir proteinler önce iki boyutlu elektroforez ile pI değerlerine ve molekül ağırlıklarına göre ayrılır ve daha sonra jelden kesilen proteinler triptik parçalanmayı takiben peptit kütle parmak izinden tanımlanırlar. Proteomik, içsel durumlara, dışarıdan gelen uyarılara ve gelişimsel değişikliklere cevap olarak hücre içindeki protein değişikliklerini saptamak için kullanılan bir tekniktir. Beyin bölgelerinin proteomik olarak incelenmesi nörodejeneratif hastalıkların kompleks yapısının anlaşılması, merkezi sinir sistemi hastalıklarının araştırılması ve bunların erken teşhisini sağlayacak biyobelirteçlerin bulunması için önemlidir. Proteomik teknolojisinin avantajı belirli bir zamanda bir doku veya hücredeki tüm proteinlerin, translasyon sonrası modifikasyonları ile birlikte, kalitatif ve kantitatif olarak tayin edilebilmesidir. Bu çalışmada absans epilepsi genetik sıçan modelinde “Genetic Absence Epilepsy Rats from Strasbourg; (GAERS)” protein ekspresyonları proteomik teknolojisi kullanılarak incelenmiştir. Bu amaçla, absans nöbetlerinin oluşumundan sorumlu beyin bölgeleri olan parietal korteks ve talamus dokuları GAERS ve kontrol sıçan beyinlerinden çıkarılmıştır. Doku örneklerindeki proteinler iki boyutlu jel elektroforezi ile ayrılmıştır. Kontrol ve GAERS gruplarında anlamlı olarak ekspresyon farklılığı gösteren protein spotları PDQuest iki boyutlu jel analiz yazılımı ile belirlenmiş ve nano-LC-ESI-MS/MS ile tanımlanmıştır. Tanımlanan proteinler korteksde ATP sentaz delta alt ünitesi ve 14-3-3 zeta izoformu, talamusda ise myelin basic protein ve makrofaj göçünü engelleyici faktördür. Tanımlanmış olan proteinlerden 14-3-3 zeta ve myelin basic proteinin jel üzerindeki varlığı western blotlama ve immüno-tanımlama yöntemi ile doğrulanmıştır. Tüm proteinlerin ekspresyonları GAERS grubunda artış göstermektedir. ABSTRACT PROFILING OF THALAMUS AND CORTEX PROTEOMES IN GENETIC ABSENCE EPILEPSY (GAERS) EXPERIMENTAL MODEL Epilepsy is a chronic disorder characterized by repeated seizures resulting from abnormal activation of neurons in the brain. It is a common serious neurological condition, with prevalence rate 10-fold higher than of multiple sclerosis that affects more than 0.5% of the world population. Absence epilepsy is a particular epileptic syndrome in which patients show generalized non-convulsive seizures characterized by a brief unresponsiveness to environmental stimuli and cessation of activity. Despite of a long time study in epilepsy, the molecular mechanisms leading to epilepsy are poorly understood. Although studies defined mutations in genes related to Na+, K+, Ca2+ channel proteins, there are few studies showing intracellular protein changes. Proteomics technique which is based on a combination of electrophoresis and mass spectrometry is a universal technique in which solubilized proteins are first separated by two-dimensional electrophoresis on the basis of their pI’s and molecular sizes and then identified by peptide mass fingerprinting following excision from the gel and tryptic digestion. This technique enables one to find protein changes in cells responding to internal states, external stimulations or developmental changes. Proteomic analysis of brain compartments may be useful to understand the complexity of neurodegenerative diseases, by investigating disorders of the central nervous system and searching for the corresponding early markers. The advantage of proteomics technology is its ability of high throughput quantitative and qualitative detection of all the proteins in cells or tissues with their post-translational modifications in a given time. In this study we investigated soluble protein expressions in a genetic rat model of absence epilepsy “Genetic Absence Epilepsy Rats from Strasbourg; (GAERS)” with proteomics technology. For this, parietal cortex and thalamus which are the brain regions responsible for the generation of absence seizures were dissected from GAERS and non-epileptic control rat brain. Proteins from each tissue sample were isolated and separated by two dimensional gel electrophoresis. Spots that showed significantly different level of expression between control and GAERS groups were detected by PDQuest two dimensional electrophoresis analysis software and identified by nano LC-ESI-MS/MS. Identified proteins were ATP synthase subunit delta and 14-3-3 zeta isoform in cortex and, myelin basic protein and macrophage migration inhibitory factor in thalamus. 14-3-3 zeta and myelin basic protein’s existence in 2D gels further validated by western blotting and immunodetermination. All proteins’ expression’s were upregulated in GAERS.
