Özet

The transmission of the energy to the higher levels is through the

food chains, starting from the main source plants. There is a balance

between all the livings in the ecosystem in which the plants are the first

ring of this chain. Environmental mutagens and other related substances

threaten humans and all other living organisms in an ecosystem. Plants

are exposed to various types of environmental agents such as agricultural

pesticides and plant growth regulators, or accidentally present in polluted air, soil or water. While pesticides used against harmful effects of

agricultural production affect the target organisms, they can affect the

non-target organisms through wrong and especially excessive

applications. The adverse effects of toxic agents that accumulate in

aquatic and terrestrial crops in an ecosystem and accumulate there as a

bioindicator in plants should be identified?. Plant biotests are very

important methods for evaluating the genotoxic contamination in the

environment. For quick determination and measurement of the

genotoxicity for these stress factors various molecular and biochemical

tests are required. Single cell gel electrophoresis is also known as Comet

assay that is used to investigate the cell response to several biotic and

abiotic genotoxic agents leading to oxidative DNA damage. The Comet

assay is used as a useful tool in assessing the potential of plants as source

of information on the genotoxic impact of dangerous pollutants and

sensitive sensors in ecosystems. This technique is based on the

quantification of denatured DNA fragments migrating out of the cell

nucleus during electrophoresis. Current review aims to give information

about Comet assay and its applications as a valuable tool for monitoring

DNA damage in different plant systems. The single cell gel

electrophoresis studies were performed by our group to determine the

genotoxic effects of different concentrations of pesticides, heavy metals

and different plant growth regulators used in tissue culture applications in

tobacco as model organisms, cereals (barley and wheat) and sunflower.
