Trombositlere fibrinojen ve gpIIb/IIIa antikorlarının bağlanmasına düşük dansiteli liprotein (ldl), okside olmuş düşük dansiteli lipoprotein ve yüksek dansiteli lipoprotein (hdl)’lerin etkilerinin flovsitometrik yöntemle incelenmesi

Özet:

LDL ve okside LDL trombosit yüzeyinde diğer hücrelerdeki klasik LDL reseptöründen farklı bir reseptöre bağlanarak trombosit aktivasyonunu arttırırlar. Biz çalışmamızda trombosit agragasyonunda önemli rolleri olan trombosit fibrinojeni ve GpIIb/IIIa reseptörü ile LDL, okside LDL ve HDL lipoproteinleri arasındaki ilişkiyi araştırdık. Kesikli dansite gradient ultrasantrifügasyon yöntemi ile LDL ve HDL fraksiyonları saflaştırıldı. LDL CuSO4 ile 24 saat boyunca 37°C’de okside edildi ve oksidasyon malondialdehit seviyeleri ile ölçüldü. Lipoproteinlerin elektroforetik etkisi ve lipid içeriği tayin edildi. ADP indüklü izole trombositler artan 9 farklı konsantrasyonda (7,5-400μg/ml) LDL veya ox-LDL ile inkübe edildi. Bu lipoproteinlerin son 3 konsantrasyonuna HDL ilave edildi. GpIIb/IIIa (CD41a) ve antifibrinojen expresyonları flovsitometrik olarak ölçüldü. Sonuçlar her bir trombosit başına spesifik antikor bağlanma kapasitelerine çevrildi. ADP ile aktivasyondan sonra antifibrinojen/trombosit ve CD41a/trombosit sayıları anlamlı derecede arttı ( p<0.001). LDL veya ox-LDL ile inkübasyondan sonra artan konsantrasyona bağlı olarak CD41a/trombosit sayısı anlamlı derecede azalırken (p<0.001) antifibrinojen/trombosit sayısı anlamlı derecede arttı (p<0.001). LDL veya ox-LDL ile muamele edilmiş trombositlere HDL ilave edildiğinde, HDL’nin hem LDL hem de ox-LDL etkisi ile artan fibrinojen sayılarını anlamlı derecede azalttığı gözlendi (p<0.001). Bu çalışmada izole edilen LDL ile in vitro ox-LDL’nin trombositlere fibrinojen bağlanmasını doza bağımlı olarak arttırdığını belirledik. Bunun yanında her iki lipoprotein trombositlere GpIIb/IIIa antikorunun bağlanmasını %100 inhibe etti. HDL ise LDL ve ox-LDL’nin fibrinojen bağlanmasındaki etkisini tersine çevirdi. Elde ettiğimiz bulgulara göre GpIIb/IIIa trombosit yüzeyinde LDL ve ox-LDL için bağlanma bölgesidir ancak bu lipoproteinlerin GpIIb/IIIa üzerindeki bağlanma domaini fibrinojen domaininden farklı bir bölgedir. SUMMARY LDL, actually oxidized LDL (ox-LDL) enhance platelet function via binding to its receptors on the platelet which are different from the ‘’classical’’ receptors of the nucleated cells. In our study, we have investigated the relationship between platelet fibrinogen and GpIIb/IIIa, which have crucial roles in platelet aggregation, and LDL, oxidized LDL and HDL lipoproteins. Human LDL and HDL were seperated by density gradient ultracentrifugation. LDL was oxidized with CuSO4 for 24h at 37°C and oxidation measured by malondialdehyte levels. Electrophoretic actions and lipid content of lipoproteins were detected. ADP (10µM) induced washed platelets were treated with increasing 9 different concentrations of LDL or ox-LDL (7,5 to 400µg/ml) where HDL has been added to final 3 concentrations of these lipoproteins. Expressions of GpIIb/IIIa (CD41a) and platelet bound fibrinogen (antifibrinogen) were measured by flow cytometry. Results were converted spesific antibody binding capasities per platelet. Following ADP activation, levels of antifibrinogen/platelet and CD41a/platelet increased significantly (p<0.001). After treatment with LDL or ox-LDL (7,5 to 400µg/ml), levels of CD41a/platelet decreased significantly whereas levels of antifibrinogen/platelet increased significantly in dose dependent manner (p<0.001 respectively). However, addition of HDL inhibited the increase in bound fibrinogen molecules (p<0.001). These data showed that while LDL and ox-LDL enhanced fibrinogen binding to platelets, increasing concentrations of LDL and ox-LDL abolished the binding of antiGpIIb/IIIa to platelets dose dependently. HDL reversed this effect of LDL or ox-LDL on only fibrinogen binding. We concluded that GpIIb/IIIa might be receptor for LDL and ox-LDL, and it seems that binding site of these lipoproteins on GpIIb/IIIa differ from fibrinogen domain.
