Stres oluşturulan sıçanlarda gelişen mesane hasarı üzerine oksitosinin etkisi: oksidatif stres ve mast hücreleri

Özet:

Bu çalışmada sıçanlarda, bir kaygı stres modeli olan sudan kaçınma stresi ile oluşturulan interstisyel sistit modelinde oksitosin tedavisinin etkinliği araştırılmıştır. Çalışmaya dahil edilen Wistar sıçanlar 5 gruba ayrıldı ve her grupta eşit sayıda erkek (n=6) ve dişi (n=6) sıçan alındı. Grup 1 (kontrol): herhangi bir işlem uygulanmadı, Grup 2 (oksitosin): 5 gün sc. 5µg /kg oksitosin uygulandı, Grup 3 (kronik stres): 5 gün günde 2 saat sudan kaçınma stresi (SKS) uygulandı, Grup 4 (kronik stres + iyileşme): kronik stresi takiben 5 gün boyunca strese maruz kalmadan beslendi ve Grup 5 (Kronik Stres + Oksitosin): kronik stres boyunca 5µg /kg oksitosin sc. uygulandı. Deney sonunda mesane dokusu ışık ve geçirimli elektron mikroskobunda incelendi. Mesane kaveolin-1 ekspresyonu ve lokalizasyonu immünohistokimyasal ve Western blot analizi ile değerlendirildi. Oksidatif stres, malondialdehit ve glutatyon ölçümleri ile değerlendirildi. Dişi ve erkek kronik stres grubunda serum kortizol değerleri ve oksidatif stres belirteçlerinde, kontrole kıyasla istatiksel olarak anlamlı derecede artarken, oksitosin ile tedavi edilen grupta istatiksel olarak anlamlı azalma tespit edilmiştir. Dişi kronik stres gruba ait granüle ve degranüle mast hücre sayısı kontrole kıyasla istatiksel olarak anlamlı artarken, oksitosin ile tedavi edilen grupta ise anlamı derecede azalmıştır. Tüm gruplara ait kaveolin-1 ekspresyonunda ve western blot analizinde anlamlı bir farklılık tespit edilmemiştir. Çalışmamızda elde edilen histolojik bulgular, mesane dokusunda kronik stres ile ortaya çıkan dejeneratif değişikliklerin, oksitosin uygulaması ile hem dişi hem de erkek sıçanlarda sınırlandığını göstermektedir. SUMMARY The Effect of Oxytocin on Stress Induced Urinary Bladder Injury: Oxidative Stress and Mast Cells In this study, we aimed to study the impact of oxytocin therapy in the water awoidance stress model in rats. Sixty Wistar rats were assigned into 5 groups and each group included the same numbers of male (n=6) and female (n=6) rats. Group 1 (control): had no intervention, Group 2 (oxytocin): recieved 5µg /kg oxytocin sc. for 5 days, Group 3 (chronic stress): underwent 2 hour daily water avoidance stress (WAS) for 5 days, Group 4 (chronic stress+recovery): underwent chronic stress and followed up without stress for the next 5 days and Group 5 (chronic stress+oxytocin): recieved 5µg /kg oxytocin sc. during chronic stress. Urinary bladder tissues were examined under light and electron microscopy. Expression and localization of caveolin-1 in urinary bladder were determined by immunohistochemistry and western blot analyses. While both male and female rats in chronic stress group revealed significantly higher levels of serum cortisole and markers of oxidative stress when compared to the controls, chronic stress + oxytocin group revealed significantly lower levels of serum cortisole and markers of oxidative stress when compared to the chronic stress. As the number of both granülated and degranülated mast cells in female rats of the chronic stress group were prominently increased with respect to the controls, it was significantly decreased in the female rats of chronic stress+oxytocin group. Expression and localization of caveolin-1showed no significance amoung the groups. Histological results of our study revealed that oxytocin application limits the degenerative alterations related to chronic stress in both male and female rats.
