Tokoferil fosfatın aterosklerozdaki rolünün in vitro in vivo sistemlerde incelenmesi

Özet:

-Tokoferol vitamin E’nin aktif olan formudur. Sinyal ileti yolakları, hücre proliferasyonu ve gen ekspresyonu dahil olmak üzere birçok hücresel olayın -tokoferol tarafından düzenlendiği gösterilmiştir. -Tokoferolün bu etkileri ateroskleroz ve inflamatuar hastalıklarda koruyucu özelliğinin temelini oluşturmaktadır. Son zamanlarda yapılan araştırmalarda -tokoferolün kroman halkasındaki hidroksil grubundan fosforile olmuş bir türevi olan tokoferil fosfatın dokularda ve besin maddelerinde doğal olarak bulunduğunun tespit edilmesi, vitamin E’nin suda çözünen bu formunun -tokoferolün doğal bir türevi olduğunu göstermektedir. Tokoferil fosfatın biyolojik etkileri ve moleküler özellikleri bilinmemektedir. Bu çalışmamızın amacı, tokoferil fosfatın etkilerinin in vitro ve in vivo ortamda araştırılmasıdır. In vitro da, tokoferil fosfatın hücre proliferasyonu ve CD36 mRNA ekspresyonu üzerindeki etkileri incelenmiş ve -tokoferol ile meydana gelen etki ile kıyaslanmıştır. Tokoferil fosfatın, sıçan aortik düz kas hücreleri ve THP-1 monosit hücrelerinde hücre proliferasyonunu, insan koroner arter düz kas hücrelerinde, CD36 mRNA ekspresyonunu -tokoferole kıyasla daha etkili bir şekilde baskıladığı gözlenmiştir. Mast hücrelerinde ve insan koroner arter düz kas hücrelerinde -tokoferolden tokoferil fosfatın sentez edildiği gözlenmiştir. Alkalen fosfatazın tokoferil fosfatı hidroliz edebildiği bulunmuştur. In vivo da, % 2 kolesterol içeren diyet ile indüklenen aterosklerotik tavşan modelinde tokoferil fosfatın ateroskleroz gelişimi üzerindeki etkisi incelenmiş, tokoferil asetat ile meydana getirilen etki ile kıyaslanmıştır. Tokoferil fosfatın aterosklerotik plak gelişiminin önlenmesinde ve CD36 mRNA ekspresyonunun baskılanmasında tokoferil asetata göre daha etkili olduğu gözlenmiştir. Tokoferil fosfatın hücresel olayların düzenlenmesinde -tokoferole göre daha etkili olduğunun saptanması, hücre içi sentezi ve hidrolizi, tokoferil fosfatın sinyal iletiminde rol oynayan bir molekül olabileceğini düşündürmektedir. SUMMARY -Tocopherol is the active form of vitamin E. It has been shown that -tocopherol regulates a number of cell properties, including signal transduction, cell proliferation and gene expression. Modulation of cellular effects is at the basis of the preventive effect of -tocopherol against atherosclerosis and inflammatory disease. Recently, a derivative of -tocopherol, tocopheryl phosphate, which is phosphorylated at the OH group of the chromane ring, has been found to be present in biological tissues and foodstuffs, indicating that this water soluble form is a natural derivative of -tocopherol. Molecular properties and biological effects of tocopheryl phosphate are not known. The aim of this work is to study the effects of tocopheryl phosphate in vitro and in vivo systems. The effects of tocopheryl phosphate on cell proliferation and CD36 mRNA expression were investigated and compared with the effect produced by -tocopherol. It has been shown that tocopheryl phosphate is more potent than -tocopherol in inhibition of cell proliferation in rat aortic smooth muscle cells and THP-1 monocyte cells. It was observed that in human coronary artery smooth muscle cells and mast cells, tocopheryl phosphate can be synthesized from -tocopherol. It was shown that alkalen phosphatase is capable of hydrolyzing tocoheryl phosphate. In vivo, the effects of tocopheryl phosphate on a rabbit model of atherosclerosis which was induced with a diet containing 2% cholesterol was investigated and compared with the effect produced by tocopheryl acetate. The results show that the atherosclerotic-preventing effect of the tocopheryl phosphate was more pronounced than that of tocopheryl acetate. The finding that tocopheryl phosphate is more potent than -tocopherol itself, it can be synthesized and hydrolyzed supports the hypothesis that tocopheryl phosphate might be a signaling molecule.
