Özet:

Büyük endüstriyel önemi göz önünde bulundurularak Van Gölü, Türkiye ‘den izole edilen mikroorganizmalar ile alkalen proteaz enziminin üretimi bu çalışma kapsamında araştırılmıştır. Morfolojik incelemeler alkalifilik 24 izolatın hepsinin çubuk şekilli (Bacilli) olduğunu göstermektedir. Her bir izolatın biyokimyasal profilinin oluşturulmasında otomatik mikrobiyal tanılama sistemi kullanılmıştır. Halotolerant izolat BA17 yalnızca geniş bir tuzluluk aralığında (0-20% NaCl) büyüme ve enzim üretim yeteneğinden dolayı değil hem de spesifik proteaz üreticisi olarak bilinen Bacillus sp. nin bir üyesi olarak tanılanması gerçeği yüzünden kaynak mikroorganizma olarak seçilmiştir. 16S rDNA temelli moleküler tanılama çalışmaları BA17 izolatının Bacillus licheniformis türüne ait olduğu göstermektedir. Büyüme besi yerini optimize etmek amacıyla farklı karbon kaynaklarının, organik ve inorganik azot kaynaklarının ve Ca+ iyonu varlığının alkalen proteaz enziminin üretimi üzerindeki etkisi incelenmiştir. Ham enzimin pH ve sıcaklık profilleri, B. licheniformis BA17 tarafından üretilen alkalen proteaz enziminin 50ºC ve pH 10.0 optimum aktvite şartları ile çeşitli endüstiyel uygulamalar için uygun bir aday olarak düşünülebileceğini göstermektedir.Bacillus licheniformis BA17 soyunun biyoreaktör kültürünün; biyokütle oluşumu, maltoz kullanımı ve alkalen proteaz aktivite profilleri gibi önemli fermentasyon parametreleri de araştırılmıştır. ABSTRACT Considering their enormous industrial importance, production of alkaline protease enzyme by microorganisms isolated from Van Lake, Turkey was investigated within the scope of this study. Morphological examination showed that all of the 24 alkaliphilic isolates were rod-shaped (Bacilli). An automated microbial identification system was used to generate the biochemical profile of each isolate. The halotolerant isolate BA17 was chosen as the source microorganism not only because of its capability of growth and enzyme production at a wide range of salinity (0-20% NaCl) but also because of the fact that it was identified as a member of the Bacillus sp., which are well-known as specific protease producers. 16S rDNA-based molecular identification studies showed that the isolate BA17 belonged to the species Bacillus licheniformis. In order to optimize the growth medium, the effect of different carbon sources, organic and inorganic nitrogen sources and presence of Ca+ ion on the production of alkaline protease enzyme was studied. pH and temperature profiles of the crude enzyme preparations revealed that the alkaline protease enzyme produced by B. licheniformis BA17 with optimum activity conditions of 50ºC and pH 10.0 could be considered as a suitable candidate for various industrial applications. Important fermentation parameters such as biomass formation, maltose utilization and alkaline protease activity profiles of bioreactor cultures of Bacillus licheniformis BA17 strain were also investigated.
